We have previously shown that copper ions are required to nitrosylate thiol groups in the core protein of glypican-1, a heparan sulfate-substituted proteoglycan. When S-nitrosylated glypican-1 is then exposed to an appropriate reducing agent, such as ascorbate, nitric oxide is released and autocatalyzes deaminative cleavage of the glypican-1 heparan sulfate side chains at sites where the glucosamines are N-unsubstituted. These processes take place in a stepwise manner, whereas glypican-1 recycles via a caveolin-1-associated pathway where copper ions could be provided by the prion protein. Here we show, by using both biochemical and microscopic techniques, that (a) the glypican-1 core protein binds copper(II) ions, reduces them to copper(I) when the thiols are nitrosylated and reoxidizes copper(I) to copper(II) when ascorbate releases nitric oxide; (b) maximally S-nitrosylated glypican-1 can cleave its own heparan sulfate chains at all available sites in a nitroxyl ion-dependent reaction; (c) free zinc(II) ions, which are redox inert, also support autocleavage of glypican-1 heparan sulfate, probably via transnitrosation, whereas they inhibit copper(II)-supported degradation; and (d) copper(II)-loaded but not zinc(II)-loaded prion protein or amyloid beta peptide support heparan sulfate degradation. As glypican-1 in prion null cells is poorly S-nitrosylated and as ectopic expression of cellular prion protein restores S-nitrosylation of glypican-1 in these cells, we propose that one function of the cellular prion protein is to deliver copper(II) for the S-nitrosylation of recycling glypican-1.","tracks":[{"project":"sentences","denotations":[{"id":"T1","span":{"begin":0,"end":132},"obj":"Sentence"},{"id":"T2","span":{"begin":133,"end":218},"obj":"Sentence"},{"id":"T3","span":{"begin":219,"end":380},"obj":"Sentence"},{"id":"T4","span":{"begin":381,"end":643},"obj":"Sentence"},{"id":"T5","span":{"begin":644,"end":814},"obj":"Sentence"},{"id":"T6","span":{"begin":815,"end":1536},"obj":"Sentence"},{"id":"T7","span":{"begin":1537,"end":1835},"obj":"Sentence"}],"namespaces":[{"prefix":"_base","uri":"http://pubannotation.org/ontology/tao.owl#"}],"attributes":[{"subj":"T1","pred":"source","obj":"sentences"},{"subj":"T2","pred":"source","obj":"sentences"},{"subj":"T3","pred":"source","obj":"sentences"},{"subj":"T4","pred":"source","obj":"sentences"},{"subj":"T5","pred":"source","obj":"sentences"},{"subj":"T6","pred":"source","obj":"sentences"},{"subj":"T7","pred":"source","obj":"sentences"}]},{"project":"Glycosmos6-GlycoEpitope","denotations":[{"id":"T1","span":{"begin":116,"end":131},"obj":"http://www.glycoepitope.jp/epitopes/EP0086"},{"id":"T2","span":{"begin":339,"end":354},"obj":"http://www.glycoepitope.jp/epitopes/EP0086"},{"id":"T3","span":{"begin":563,"end":578},"obj":"http://www.glycoepitope.jp/epitopes/EP0086"},{"id":"T4","span":{"begin":1138,"end":1153},"obj":"http://www.glycoepitope.jp/epitopes/EP0086"},{"id":"T5","span":{"begin":1309,"end":1324},"obj":"http://www.glycoepitope.jp/epitopes/EP0086"},{"id":"T6","span":{"begin":1508,"end":1523},"obj":"http://www.glycoepitope.jp/epitopes/EP0086"}],"attributes":[{"subj":"T1","pred":"source","obj":"Glycosmos6-GlycoEpitope"},{"subj":"T2","pred":"source","obj":"Glycosmos6-GlycoEpitope"},{"subj":"T3","pred":"source","obj":"Glycosmos6-GlycoEpitope"},{"subj":"T4","pred":"source","obj":"Glycosmos6-GlycoEpitope"},{"subj":"T5","pred":"source","obj":"Glycosmos6-GlycoEpitope"},{"subj":"T6","pred":"source","obj":"Glycosmos6-GlycoEpitope"}]},{"project":"mondo_disease","denotations":[{"id":"T1","span":{"begin":7,"end":14},"obj":"Disease"},{"id":"T2","span":{"begin":196,"end":203},"obj":"Disease"},{"id":"T3","span":{"begin":1479,"end":1486},"obj":"Disease"}],"attributes":[{"id":"A1","pred":"mondo_id","subj":"T1","obj":"http://purl.obolibrary.org/obo/MONDO_0019065"},{"id":"A2","pred":"mondo_id","subj":"T2","obj":"http://purl.obolibrary.org/obo/MONDO_0019065"},{"id":"A3","pred":"mondo_id","subj":"T3","obj":"http://purl.obolibrary.org/obo/MONDO_0019065"},{"subj":"T1","pred":"source","obj":"mondo_disease"},{"subj":"T2","pred":"source","obj":"mondo_disease"},{"subj":"T3","pred":"source","obj":"mondo_disease"}]},{"project":"CL-cell","denotations":[{"id":"T1","span":{"begin":1560,"end":1570},"obj":"Cell"}],"attributes":[{"id":"A1","pred":"cl_id","subj":"T1","obj":"http://purl.obolibrary.org/obo/CL:0000623"},{"id":"A2","pred":"cl_id","subj":"T1","obj":"http://purl.obolibrary.org/obo/CL:0000825"},{"subj":"T1","pred":"source","obj":"CL-cell"}]}],"config":{"attribute types":[{"pred":"source","value type":"selection","values":[{"id":"sentences","color":"#93a5ec","default":true},{"id":"Glycosmos6-GlycoEpitope","color":"#bfec93"},{"id":"mondo_disease","color":"#ec93d9"},{"id":"CL-cell","color":"#93ece5"}]}]}}
