The RNA was treated with DNase I (QIAGEN, Venlo, Netherlands). Then first-strand cDNA was synthesized using a PrimeScript RT reagent Kit (Takara Bio, Otsu, Japan). We carried out real-time quantitative RT-PCR for the mRNA expression of secreted phosphoprotein1 (Spp1) in the renal cortex by using a 7900HT Fast Real-Time PCR System with commercial TaqMan probe kits (TaqMan Gene Expression Assays, (Life Technologies) and TaqMan Universal PCR Master Mix (Life Technologies) according to a standard method.
