To confirm that the eIF4G is involved, we took another approach by treating cells with Trail. It has been reported that treating HeLa cells with Trail for 4 h would cause cleavage of eIF4G, thus, impair the cap-dependent translation initiation and decrease the incorporation of [35S]methionine (32). To test the effect of Trail on the expression of M2, we treated HeLa cells with Trail for 4 h and then detected the protein level of M2 using western blot. As shown in Figure 8A, the endogenous M2 protein level was increased following treatment with Trail. However, the M2 mRNA level was not changed significantly as determined by RNase protection assay (Figure 8B). The result of pulse-labeling by [35S]methionine and immunoprecipitation following Trail treatment showed that the synthesis rate of M2 protein was increased (Figure 8C). Hence, the increase in the level of endogenous M2 protein following Trail treatment is likely due to the increased synthesis rate of M2 protein and it may be by activating the translation of M2 transcripts with long 5′-UTRs.
